LSP Optimal Kinase Library

This is a collection of characterized kinase inhibitors purchased from a variety of vendors and provided to ICCB-Longwood by HMS LSP. Each compound is present at 4 concentrations (10 mM, 1 mM, 0.1 mM and 0.01 mM). It was plated in January 2021.

The LSP Optimal Kinase Library is designed to probe the kinome with the most selective compounds as well as small molecule drugs and compounds in clinical development. Extended annotation on the targets of these compounds and information on how these compounds relate to each other in phenotypic fingerprint, Target Affinity Spectrum and structure is available via www.smallmoleculesuite.org.

This library is designed to have two inhibitors that are non-similar in structure, but comparable in selectivity for each kinase as described in PMCID:PMC6526536. The advantage of having two structurally distinct compounds is that they are more likely to have orthogonal target affinity spectra (TAS) (i.e. different off-target affinities). This feature enables users with a way to check if an observed effect is likely due to on-target or off-target action of the molecule.
