ICCB-Longwood Mechanism of Action Library 10mM

The ICCB-Longwood Mechanism of Action Library (ICCB-L MoA) is a growing collection of well-annotated, high quality compounds with known biological targets. A tool score, described by Wang and colleagues (Cell Chemical Biology 23, 862-874), was utilized to identify the best target-compound pairs based upon target specificity and potency, with Rank 1 being the highest. This collection, when complete, will also include Rank 2 and Rank 3 compounds for some biological targets.

The curation of this library was made possible through the support of HMS LINCS and guidance from Yuan Wang and Jeremy Jenkins at Novartis Institutes for BioMedical Research. Compounds were purchased by ICCB-Longwood and the Assay Development & Screening Facility at Boston Children's Hospital, and funded in part by a grant to BCH from Massachusetts Life Sciences Center. Harvard Medical School's Foundry Award Program also provided funds for the acquisition of this library.

Note that two wells contain fluorescein (3744:C17 and 3752:I07), a highly fluorescent dye with a bright yellowish-green emission that may produce false results in fluorescence and absorbance assays.

Each compound is plated at 10 mM in Quadrant A1 of the 384-well plate. This library is primarily utilized by investigators screening in 96-well format. Please see map below for a visual layout.

Plates 3740-3744 were plated in October 2017 and contain 400 Rank 1 compounds. Plates 3745-3748 were plated in February 2018 and contain 320 Rank 1 and 2 compounds. Plates 3749 and 3750 were plated in September 2018 and contain Rank 1, 2 and 3 compounds. Plates 3751-3755 were plated in October 2020 and contain Rank 1, 2 and 3 compounds.
