# RNA Transfection

ICCB-Longwood uses an integrated linear platform that includes the Agilent 4-R BenchCel, Bravo and PlateLoc. This setup, contained in a Baker BioProtect II biosafety cabinet, and the associated Thermo Multidrop Combi provides a precise and flexible platform allowing for a wide array of applications.

- Just-in-time library delivery during the transfection step of sgRNA/siRNA/miRNA screens
- Approximately three minutes to transfer contents from one library plate to three assay plates
- Automate adding assay-specific controls
- Reformat libraries
- Custom ICCB-L projects

## Agilent Bravo Automated Pipettor

- 96- and 384-well heads enable transfer of RNA reagents into 96- and 384-well microplates
- Disposable tips are used to eliminate the potential for cross contamination (available from Agilent or Fluotics Ltd.)
- A program is written and calibrated for each transfer, as the volume and number of replicates varies by screen
- Typical transfer volumes range between 1 – 3 uL per replicate
- An ‘intermediate plate’ is used when transferring to more than 3 assay plates

## Agilent 4-R BenchCel Plate Handler

Delivers large numbers of library plates and tip boxes to and from the Bravo deck for processing

## Agilent PlateLoc Heat Sealer

Quickly and effectively seals library plates after the transfer
