#  ICCB-Longwood Mechanism of Action Library 

 



The ICCB-Longwood Mechanism of Action Library (ICCB-L MoA) is a growing collection of well-annotated, high quality compounds with known biological targets. A tool score, described by Wang and colleagues ([Cell Chemical Biology 23, 862-874](https://www.ncbi.nlm.nih.gov/pubmed/27427232)), was utilized to identify the best target-compound pairs based upon target specificity and potency, with Rank 1 being the highest. This collection, when complete, will also include Rank 2 and Rank 3 compounds for some biological targets.

The curation of this library was made possible through the support of [HMS LINCS](http://lincs.hms.harvard.edu/) and guidance from Yuan Wang and Jeremy Jenkins at Novartis Institutes for BioMedical Research. Compounds were purchased by ICCB-Longwood and the [Assay Development & Screening Facility](https://www.childrenshospital.org/node/152965/cores/assay-development-and-screening-facility) at Boston Children's Hospital, and funded in part by a grant to BCH from [Massachusetts Life Sciences Center](http://www.masslifesciences.com/). Harvard Medical School's [Foundry Award Program](https://foundryawards.hms.harvard.edu/) also provided funds for the acquisition of this library.

Note that 8 wells contain fluorescein (3724:C17, 3724:C18, 3724:D17, 3724:D18, 3732:I07, 3732:I08, 3732:J07, and 3732:J08 ), a highly fluorescent dye with a bright yellowish-green emission that may produce false results in fluorescence and absorbance assays.

Each compound is plated at 4 concentrations (10 mM, ~2 mM, ~0.4 mM and ~0.08 mM). The concentrations of the compounds on this library plate are according to quadrant; Quadrant A1 is at 10 mM, Quadrant A2 is at ~2 mM, Quadrant B1 is at ~0.4 mM and Quadrant B2 is at ~0.08 mM, please see map below for a visual layout.

Plates 3720-3724 were plated in October 2017 and contain 400 Rank 1 compounds. Plates 3725-3728 were plated in February 2018 and contain 320 Rank 1 and 2 compounds. Plates 3729 and 3730 were plated in September 2018 and contain Rank 1, 2 and 3 compounds. Plates 3731-3735 were plated in October 2020 and contain Rank 1, 2 and 3 compounds.
